I asked my son to mail a box at the post office in my neighborhood. He mailed the box to my sister in Atlanta on June 1st and it never arrived. My return address is also on the box. My son has lost the receipt so I have no tracking number. I put together this box of clothing and handbags for my sister. She was down and out and I wanted to surprise her. I'm really at a loss as to why it didn't get there or come back to me.

You will need to go to the post office where mailed and talk to the postmaster or whoever is in charge and give them as much information as possible so they might be able to look for your package.
IT MAY HAVE BEEN DELIVERED TO SOMEONE ELSE WHO DECIDED TO KEEP IT.TALK TO YOUR POST OFFICE THEY MAY BE ABLE TO TRACK YOUR PACKAGE.
I am so sorry to hear that you are dealing with a lost package. It is very frustrating! Just last week I had a package return to sender (to me) from 6 weeks ago...I was livid as it was something I sold and the person was going to give me negative feedback because the USPS tracking site only said in transit.

The tracking number is going to be key to the process so if you postmaster (talk to him or her, not just someone who works at the counter if you can) can find that, it will help. Once you have that, the USPS can pinpoint EXACTLY where the package is in their system.
The one thing I would add is in parallel, is talk to your sister to see if anyone in her area has cameras on their homes/businesses and record the activity and if she could look at the video from the week span when the package was to have been delivered.
Where I am we recently had a spate of people following both the postman and the white Amazon vans and helping themselves to items left on porches and mailboxes. In at least one case, they were found by people who had video of it. Yeah technology in this case!
If you go through the lost package process and it still isn't found, you can go through a second USPS process to file an insurance claim for the item IF it was sent with insurance (it comes automatically with it for priority mail up to $50).
When you are filing that process it helps to have and give them (scan to PDF) all the info you can...including if you have the store receipts for the items that were lost (even if it is just your credit card bill--BLACK OUT OTHER ITEMS AND THE LAST 4 OF THE CARD), scan them. If you have a store ad with the picture of the item, scan that also. If you bought it online, do a screen print of your purchase and make sure to capture a picture of the item.
The post office is protective of what is essentially our tax dollars so they do not issue insurance claim pay outs willy nilly...as they want as much proof as possible that you really did have (for example) a Dooney purse in the bag and not just a dime store bag. The more info you give them as evidence to show EXACTLY what was lost, better the chance of having the claim approved.
When asking for a refund, it should be the total cost of the items + the shipping cost.

People steal packages off porches now. If you have no tracking number, it will be difficult to find out if the package ever made it to the house at all. In the future, when you send a package, instruct the post office not to deliver the actual package, but to send a notice instructing the person to pick it up at the post office.
Thank you. Yes my sister has instructed them in the past NOT to drop the actual package but to leave a notification slip on her door.
