Unless you have spilled bleach on your skirt this could very likely be a soap residue spot on your skirt.
You will need white vinegar and an old toothbrush.
Step 1
Fill your sink with cold water and a 1/4 cup of white vinegar.
Step 2
Soak the garment in the water.
Step 3
Use your hands and the old toothbrush to work on the stain.
Step 4
This should loosen the soap and remove the stain.
Step 5
If this stain wasn't from washing and it isn't soap then we need to determine what the stain is from.

There are several other possibilities why you have this stain on your clothes.

White wine, milk or baby formula, bird accident, or correctional fluids.

Each one of these stains has a different way to remove them. If the vinegar and water didn't remove your stain I would suggest trying one of these other methods.
Lay the garment on a flat surface. Place a piece of paper towel under the stained area to prevent any bleeding. Work 1 tablespoon of liquid laundry detergent into the stain with a soft-bristled brush. Allow the soap to absorb into the stain for five minutes.

Sprinkle corn starch on the stain let sit 15 minutes.

Brush away corn starch until stain is absorbed.
Step 3
Put vinegar on cloth and wipe off stain.
Step 4
Leave the vinegar dry.

Then wash it away with laundry soap and water.
Do you know what made the spot? Has the skirt been dried? Is this a dark colored skirt?
Be very careful with trying different solutions as some are not intended for nylon and may not work properly.
Do not dry until you are certain the spot has been removed.
